In the present study, we have investigated the biosynthesis of dextransucrases by two strains of Leuconostoc mesenteroides isolated from Bulgarian fermented products. Extracellular dextransucrase activities of 10.80 U/ml for strain Lm 28 and 9.6 U/ml for strain Lm 22 were measured in a batch culture. The SDS-PAGE analysis showed that the molecular weight of the dextransucrases isolated from both strains was 180 kDa. Enzymes of the studied strains were found to efficiently transfer the glucosyl moiety of sucrose onto maltose acceptor. By increasing the sucrose/maltose ratio, it was possible to catalyze the synthesis of oligosaccharides of increasing degree of polymerization.

Dextransucrase production by Leuconostoc mesenteroides NRRL B-512F in media containing carbon sources other than sucrose is reported for the first time. Dextransucrases were analyzed by gel electrophoresis and by an in situ activity assay. Their polymers and acceptor reaction products were also compared by (13)C nuclear magnetic resonance and high-performance liquid chromatography techniques, r...

Comparison of exopolysaccharide production by indigenous Leuconostoc mesenteroides strains in whey medium

Two Leuconostoc mesenteroides strains NCDC 744 and NCDC 745 were evaluated for exopolysaccharides (EPS) production in whey and MRS medium. Paneer whey medium supplemented with 10% sucrose, 0.1 % yeast extract and 0.1 % K2HPO4 was found to be proferred medium for EPS production. Among two strains L. mesenteroides NCDC 744 produced significantly higher EPS (12.7 ± 0.24 gm/L) as compared to NCDC 7...

Characterization of the Dextran Produced by Leuconostoc mesenteroides from Date Fruit Extract

In the present study, the production of dextran from date extract and sucrose as carbon sources by the bacterium Leuconostoc mesenteroides NRRL B512 (f) was investigated. In comparison to blue dextran (Mw≈2000 Da), dextran molecular weight was reduced (Mw2000 KDa). Flow behavior indices of dextran solutions from date extracts were investigated and rheological parameters were evaluated at concen...

Production of Dextran by Newly Isolated Strains of Leuconostoc mesenteroides PCSIR-4 and PCSIR-9

Different isolated strains of Leuconostoc mesenteroides produce dextran compared with that of L. mesenteroides NRRL B-512 F. Among nine newly isolated strains, strains PCSIR-4 and PCSIR-9 produced dextran of different quality. Using these two newly isolated strains, the development of dextran coincided closely with the disappearance of sucrose. The yield of dextran increased during growth and m...
